In situ sequencing (ISS) is a spatial transcriptomic method to analyse gene expression in large tissue sections at single cell resolution (1, 2, 3). Using combinatorial barcoding, ISS can identify up to ~300 genes simultaneously, being an ideal tool for the discovery of new cell types, understanding cell-cell heterogeneity, generation of single cell atlases, and more.
